Chrome extension shortcuts normally work only when Chrome has focus. Use pause-only on the shortcut before you unmute, and keep volume low with the scroll wheel in between.
Chrome lets you change a command scope to Global, which makes it respond even when Chrome is not the active window. The extension declares four commands, and each can be set to Global at chrome://extensions/shortcuts. The default state is not global, because that is a choice Chrome leaves to you, so enable it for the commands you want.

- Visit chrome://extensions/shortcuts.
- Find Spotify-Web Playback Controller and open each command.
- Set the scope dropdown to Global for the commands you want system-wide.
